He's an amazing character, but his Klingon-ness made him the ultimate straight man. The prune juice bit is pretty awesome, but I think the comedic one-liner of the series had to be Worf's "I am NOT a merry man!" line in Q-Pid. One of them is when Picard, Guinan, Ro and Keiko are kids due to the anomaly (Rascals).
